SLS’ Plant gDNA Extraction Kit (Mini) provides user-friendly and easy methods for the purification of total genomic DNA from small amounts of starting samples used in various downstream processes.
This simple protocol is ideal for the simultaneous processing of multiple samples and yields pure DNA ready for direct amplification within minutes. It requires no phenol/chloroform extraction or alcohol precipitation and involves minimal handling. DNA is eluted in Buffer DE, ready for direct addition to PCR or other enzymatic reactions. Alternatively, it can be safely stored at –20°C for later use. The purified DNA is free from proteins, nucleases, contaminants, and inhibitors.
Principle
Cells are lysed during a short incubation in the presence of chaotropic salts and detergents, which immediately inactivate all nucleases. Cellular nucleic acids are then allowed to bind selectively to a glass-fibre membrane in a special centrifuge tube. The nucleic acids remain bound while a series of rapid wash-and-spin steps removes contaminating small molecules. Finally, low-salt elution removes the nucleic acids from the glass-fibre membrane.
The process does not require organic solvent extraction or extensive handling of the nucleic acids.
